After oral administration of molnupiravir, NHC circulates systemically and is phosphorylated intracellularly to NHC triphosphate. NHC triphosphate is incorporated into viral RNA by viral RNA polymerase and subsequently misdirects the viral polymerase to incorporate either guanosine or adenosine during viral replication. This leads to an accumulation of deleterious errors throughout the viral genome that ultimately render the virus noninfectious and unable to replicate.
